Abstract
<jats:p> This work represents the first systematic analysis of kinetochore assembly in human mitotic cell extracts. Using recombinant and native proteins, we show that only a few assembly steps are permitted, suggesting that multiple mechanisms guard against the spurious formation of microtubule-binding kinetochore particles in the cytoplasm of mitotic cells. </jats:p>
